The Human body is a vast network of interacting genes, proteins, and metabolites. These components, which may be considered host factors, change under disease, treatment or healthy condition. While treatment of many diseases depends on therapeutic drugs, vaccines remain the most effective long-term public health intervention to prevent infectious diseases. To date, vaccines have been developed to treat entire populations with little provision for predisposing individual host factor differences. However, the use and application of vaccines is facing multiple challenges with increasing numbers of vaccine non-responders and vaccine-relapsed individuals. The cause of this complication is partially due to host-factors. Another challenge is the adverse effects of vaccines in patients with primary immunodeficiency or autoimmune diseases, as well as vaccine-waning immunity in ageing populations, obese populations, or those with co-infection. To overcome these challenges, the solution may be the design, and formulation of precision vaccines, which are patient-specific.

Many infectious diseases are more frequent and are associated with high morbidity and mortality in older adults. Vaccination is the most efficient strategy to prevent infections, and older adults are an important target population for vaccination in order to promote health in this age group. Age-related changes in the immune system as well as other factors, such as comorbidities, obesity or frailty, influence vaccine-induced immune responses in old age. Awareness that vaccines developed for children might not be optimal for adults, and particularly for the older population, has only arisen in the recent past. Vaccination against influenza, pneumococcal disease, and herpes zoster is specifically recommended for older adults in many countries, and various strategies have been pursued in order to optimize these vaccines. However, there are still many pathogens, which severely affect the older population, but for which no vaccines are currently available. Extensive research and development are ongoing to further improve existing vaccines and to design novel vaccines in order to provide protection for this vulnerable age group. By two years of age, healthy infants in the United States can receive up to 20 vaccinations to protect against 11 diseases. Although most people know that vaccines effectively protect against serious infectious diseases, approximately one-quarter of parents in a recent survey believe that infants get more vaccines than are good for them, and that too many immunizations could overwhelm an infant's immune system. The Immunization Safety Review Committee reviewed the evidence regarding the hypothesis that multiple immunizations increase the risk for immune dysfunction. Specifically, the committee looked at evidence of potential biological mechanisms and at epidemiological evidence for or against causality related to risk for infections, the autoimmune disease type 1 diabetes, and allergic disorders.
