Protocols in Human Molecular Genetics

Protocols in Human Molecular Genetics PDF Author: Christopher G. Mathew
Publisher: Springer Science & Business Media
ISBN: 1592594964
Category : Medical
Languages : en
Pages : 461

Book Description
Extraordinary advances have been made in the field of human molecular genetics during the past five years. The ability to amplify a specific region ofDNA a millionfold in a few hours using the polymerase chain reaction has led to the rapid identification of mutations in human disease and of DNA sequence polymorphisms on every human chro- some. DNA fragments of up to 1 megabase in length can now be resolved by pulsed-field gel electrophoresis to create long-range physical maps of important regions of the genome, and can be cloned in the form of yeast artificial chromosomes. The discovery of highly variable "minisatellite" DNA sequences has led to the development of DNA fingerprinting. The application of these techniques to the study of the human genome has culminated in major advances such as the cloning of the cystic fibrosis gene, the construction of genetic linkage maps of each human chro- some, the mapping of many genes responsible for human inherited d- orders, genetic fingerprinting of forensic specimens, and the detection of mutations involved in the development of human tumors. Although many of the new techniques in molecular genetics can be learned relatively easily, it is often difficult for a researcher to obtain all of the relevant information necessary for getting up a technique and applying it successfully. The information available in the research lite- ture often lacks the depth of description that the new user requires.

Gene Mapping, Discovery, and Expression

Gene Mapping, Discovery, and Expression PDF Author: Minou Bina
Publisher: Springer Science & Business Media
ISBN: 1597450979
Category : Science
Languages : en
Pages : 335

Book Description
Completion of the sequence of the human genome represents an unpar- leled achievement in the history of biology. The project has produced nearly complete, highly accurate, and comprehensive sequences of genomes of s- eral organisms including human, mouse, drosophila, and yeast. Furthermore, the development of high-throughput technologies has led to an explosion of projects to sequence the genomes of additional organisms including rat, chimp, dog, bee, chicken, and the list is expanding. The nearly completed draft of genomic sequences from numerous species has opened a new era of research in biology and in biomedical sciences. In keeping with the interdisciplinary nature of the new scientific era, the chapters in Gene Mapping, Discovery, and Expression: Methods and Protocols recapitulate the necessity of integration of experimental and computational tools for solving - portant research problems. The general underlying theme of this volume is DNA sequence-based technologies. At one level, the book highlights the importance of databases, genome-browsers, and web-based tools for data access and ana- sis. More specifically, sequencing projects routinely deposit their data in p- licly available databases including GenBank, at the National Center of Biotechnology (NCBI) in the United States; EMBL, maintained by the European Bioinformatics Institute; and DDBJ, the DNA Data Bank of Japan. Currently, several browsers offer facile access to numerous genomic DNA sequences for gene mapping and data retrieval.

Book Description
The general underlying theme of "Gene Mapping, Discovery, and Expression" is DNA-sequence-based technologies. At one level, the book highlights the importance of databases, genome-browsers, and web-based tools for data access and analysis. This volume also provides a computational protocol for identifying and mapping recent segmental and gene duplications, as well as step-by-step procedures for identifying paralogous genes, using the genome browser at UCSC. Lastly, several chapters in this volume offer non-invasive technologies for the isolation of transcription factor complexes formed with specific DNA sequences used as bait. These methods can lead to the discovery of novel transcription factors with important roles in the control of gene expression.

Protocols for Nucleic Acid Analysis by Nonradioactive Probes

Protocols for Nucleic Acid Analysis by Nonradioactive Probes PDF Author: Elena Hilario
Publisher: Springer Science & Business Media
ISBN: 1597452297
Category : Science
Languages : en
Pages : 318

Book Description
Protocols for Nucleic Acid Analysis by Non-radioactive Probes, Second Edition provides a firm background on the basic preparative protocols required for the analysis of nucleic acids by nonradioactive methods. Presenting the methodologies using amazing new applications, this volume offers guide chapters on nucleic acid extractions, preparation of nucleic acid blots, and labeling of nucleic acids with nonradioactive haptens. New fluorescent techniques such as Real Time PCR and microarrays are also included, allowing users to get a nonradioactive protocol implemented in the laboratory with minimum adaptation required and fastest time to results. The protocols follow the successful Methods in Molecular BiologyTM series format, each offering step-by-step laboratory instructions, an introduction outlining the principles behind the technique, lists of the necessary equipment and reagents, and tips on troubleshooting and avoiding known pitfalls.

Methods in Plant Molecular Biology and Biotechnology

Methods in Plant Molecular Biology and Biotechnology PDF Author: Bernard R. Glick
Publisher: CRC Press
ISBN: 1351082949
Category : Science
Languages : en
Pages : 382

Book Description
Methods in Plant Molecular Biology and Biotechnology emphasizes a variety of well-tested methods in plant molecular biology and biotechnology. For each detailed and tested protocol presented, a brief overview of the methodology is provided. This overview considers why the protocol is used, what other comparable methods are available, and what limitations can be expected with the protocol. Other chapters in the book present overviews regarding how to approach particular problems and introduce unique methods - such as how to use computer methodology to study isolated genes. The book will be a practical reference for plant physiologists, plant molecular biologists, phytopathologists, and microbiologists.

Nonmammalian Genomic Analysis

Nonmammalian Genomic Analysis PDF Author: Bruce Birren
Publisher: Academic Press
ISBN: 9780080537726
Category : Science
Languages : en
Pages : 353

Book Description
Offering detailed protocols for those needing to construct a variety of maps and isolate genes, this unique book is intended to popularize the new techniques of genome analysis derived from the Human Genome Project. The power of these new methods is often most striking when applied to problems outside of human genetics, particularly the nonmammalian systems on which many researchers focus. Many of these organisms are economically important and biologically rich. Nonmammalian Genomic Analysis: A Practical Guide covers the "how to" aspects of preparation, handling, cloning, and analysis of large DNA and the creation of chromosome and genome maps. This lab manual facilitates the transfer of these technologies to small "low tech" environments and allows them to be used by those with no background in genome mapping or large-fragment cloning. Like having a local expert, this collection provides procedures for anyone, anywhere, and allows the replication of others' success. Includes detailed and clearly-written step-by-step protocols Evinces expected results and offers trouble shooting advice Provides techniques appropriate for small laboratories as well as those with limited resources Covers a broad variety of cloning systems, including single copy vectors Discusses a diverse range of organisms, from prokaryotes to eukaryotes, from single-celled organisms to highly complex organisms
